    Usually in the case when the guy secretly is in love with his female friend. But she doesn't want him as a boyfriend.

    Exactly ^^ thats been my point from the start. He wouldnt want her as a best mate unless he has feelings for her (the only exception is if she looks and acts like a man)

    Of course they can, but their relationship will never be as fulfilling and wonderful as two partners who are also best friends. I know this from experience :-).
